Zum Spektrum der aktuell untersuchten Krankheitsgruppen gehören die autosomalen Gliedergürtelmuskeldystrophien ( LGMD ) und autosomal erblichen myofibrillären Myopathien ( MFM ) sowie die erworbene Einschlußkörpermyositis ( IBM ).
Derzeit führen wir im Hinblick auf die LGMD- und MFM-Formen eingehende Charakterisierungen der Filamin C-assoziierten molekularen Pathologie unter Berücksichtigung neu identifizierter Mutationen und detektierter Beeinträchtigungen des Ubiquitin-Proteasom-System (UPS) und des Autophagie-vermittelten Proteinabbaus durch.
Diese Arbeiten beinhalten Transfektionsstudien an murinen Myoblasten-Zelllinien, Charakterisierungen des mutierten Filamin C-Proteins und massenspektrometrische Analysen von mittels Laser-Mikrodissektion isolierten pathologischen Protein-Aggregaten, sowie die Identifikation und Charakterisierung neuer und krankheitsassoziierter Filamin-Bindungspartner.

With regard to the MFM and LGMD forms we currently conduct detailed characterizations of the Filamin C-associated molecular pathology with respect to newly identified mutations and detected impairments of the ubiquitin-proteasome system (UPS) and autophagy-mediated protein degradation.
These works include transfection of murine myoblast cell lines, characterization of the mutant Filamin C protein and mass spectrometric analysis by laser micro-dissection isolated pathological protein aggregates, as well as the identification and characterization of new and disease-associated Filamin-binding partners.
